The plot of the film centres on the death of Gus Portokalos, the family patriarch, and the Portokalos family's journey to Greece for a reunion in his hometown of Vrisi. Toula, Gus's daughter, attempts to locate her father's childhood friends to fulfil his wish of having his journal—containing his life story—handed over to them. The family is invited to Vrisi by Victory, a distant relative and the town's mayor, under the guise of a family reunion. Upon their arrival, they discover that the village is nearly abandoned and that Victory's true intention is to have the Portokaloses revive the village.

The family goes sightseeing and visits Gus's childhood home, where they meet Alexandra, Gus's old flame. The following day, Toula searches for Gus's friends in a nearby town but is unsuccessful and ends up getting drunk on ouzo.
