The Portokalos family is dealing with grief and loss in the third instalment of the My Big Fat Greek Wedding film series. Toula (Nia Vardalos) and Ian (John Corbett) are happily married, but facing the challenges of parenthood as their daughter Paris (Elena Kampouris) struggles with the responsibilities of college. Nick (Louis Mandylor), now separated, returns home and displays immature behaviour. Athena and Yiannis are caring for Maria (Lainie Kazan), who is battling dementia.
The family travels to Greece for a reunion in the hometown of the late Gus Portokalos, Toula's father. They are invited by Victory (Melina Kotselou), the mayor of Gus' hometown, Vrisi. On the flight, Aunt Voula (Andrea Martin) introduces her assistant Aristotle (Elias Kacavas), who is also Paris' college-mate, and Toula reveals she is bringing Gus' journal, containing his life story, to give to his three childhood friends.
Upon arriving in Athens, the family goes sightseeing and finds Gus' childhood home in the nearly abandoned Vrisi. There, they meet Alexandra (Anthi Andreopoulou), Gus's old flame. Victory confesses that there is no reunion, and she had hoped the Portokalos family would help revive the village. The next day, the family goes to a nearby town, where Toula searches for Gus's friends without success and gets drunk on ouzo.
Meanwhile, Ian befriends a monk who has information about everyone's activities in the village. At dinner, the family meets Peter (Alexis Georgoulis), Alexandra's son and Gus' unknown son, and his son Christos (Giannis Vasilottos). Christos is in a secret relationship with a Syrian refugee named Qamar (Stephanie Nur), and they plan to get engaged despite fearing their families' disapproval.
The family helps plan Christos and Qamar's wedding, which combines Greek and Syrian wedding traditions. Paris and Aristotle decide to explore their relationship further. Toula, Peter, and Nick scatter Gus' ashes at the oldest olive tree in the village.
