The film follows the Portokalos family as they travel to Greece for a family reunion and to fulfil the wish of their late father, Gus. Upon arriving in Greece, they learn that this will be harder than anticipated. They also discover that Gus had a son, Peter, with the village matriarch, Alexandra. Peter has a son, Christos, who is in a secret relationship with a Syrian refugee, Qamar, as he fears his family won't approve. When the secret is revealed, Christos and Qamar decide to get engaged, despite initial disapproval from Peter and Alexandra. The film ends with a big, fat, Greek-Syrian wedding, celebrating both Greek and Syrian wedding traditions.

In addition to the wedding, other key plot points include Paris, Toula and Ian's daughter, failing her first year of college and deciding to explore a relationship with Aristotle, her old flame. The family also buries Gus's ashes at the oldest olive tree in the village.
